Do I lose my Great Vault reward if I don't pick it up?

Yes. The vault holds one week's rewards, and when the next reset lands, the old selection is replaced by the new one. Log in, open the vault, take an item — even a piece you plan to disenchant or use for a tier slot. Making the choice costs nothing.

How many runs do I need before the vault resets?

The dungeon row unlocks at 1, 4 and 8 Mythic+ runs completed that week. The raid row unlocks at 2, 4 and 6 bosses killed. The third row tracks the season's world or PvP content. You get up to nine options total, but you pick exactly one item.

Only runs finished before the reset count. A key completed ten minutes after the rollover belongs to the new week.
