What actually resets each week?

So on a Tuesday morning in the US, the daily and weekly resets happen simultaneously - that is why servers usually go down for maintenance around that window.

Do I lose my Great Vault if I do not claim it?

How do I check the exact timer in game?

Open the Raid Info panel (the Instances tab in your Group Finder window) - every saved lockout lists the exact time it expires. The Great Vault interface in your faction hub also shows a live countdown to the next reset.
